Course objectives
At the end of the course students should be able to understand and explain these topics - Principles of chemical and physical methods used in biochemical research. Their instrumentation and use in routine analysis and study of biochemical processes. Metodological approaches in study of biomolecules behaviour and their interconversions.
Syllabus
  • 1. Introduction. Principles of the work with biological material. Strategy and planning. Desintegration of tissues and cells.
  • 2. Preparative centrifugation. Sedimentation analysis.
  • 3. Phase separations. Precipitation and extraction.
  • 4. Membrane separations. Concentration and drying. Water purification.
  • 5. Chromatographic methods. General principles and characte- ristics. Adsorption and distribution chromatographies.
  • 6. Ion exchange chromatography, chromatofocusation. Gel and affinity chromatographies. Gas chromatography.
  • 7. Mass spectrometry.
  • 8. Electromigration methods. General characteristics and side effects. Electrophoresis, its modifications. Isotachophoresis.
  • 9. Immunochemical methods.
  • 10. Spectral methods. General characteristics, classification. Electronic spectra of atoms. AES and AAS. Applications in biochemistry.
  • 11. Electronic spectra of molecules.
  • 12. UV-VIS spectroscopy, principles, use.
  • 13. Types of instruments, difference spectra, perturbation. Turbidimetry and nefelometry.
  • 14. Luminiscence methods, principles. Spectrofluorometry, principles, use.
  • 15. Biochemical applications, fluorescence polarisation, probes.
  • 16. IR and Raman spectroscopy.
  • 17. Moessbauer spectra.
  • 18. NMR, biochemical applications. EPR, biochemical applications.
  • 19. Chiroptic methods. ORD and CD. Principle, meaning and applications in biochemistry.
  • 20. Calorimetry, types of instruments, biochemical applications.
  • 21. Methods of determination of the size and shape of macromole- cules. Isotope exchange.
  • 22. Coligative methods, osmometry. Viscosimetry.
  • 23. Translational and rotational diffusion.
  • 24. Light and X-rays scattering, rentgenostructure analysis.
  • 25. Electrochemical methods in biochemistry. Principles, classification. Potentiometry, ISE. Bioselective electrodes.
  • 26. Amperometric methods. Polarography and voltammetry, bio- sensors.
  • 27. Fast reactions, metods, biochemical applications.
