Prerequisites
Following lectures is appropriate attend before or during exercise: C7021 Separation Methods A, C7031 Analytical Atomic Spectrometry, C7041 Molecular Spectrometry, C7050 Electroanalytical Methods, C7830 Capillary Electrophoresis and C7895 Mass Spectrometry of Biomolecules.

Course objectives
Main objectives can be summarized as practice training in electroanalytical methods, optical spectroscopy, mass spectroscopy, and separation methods. A) Modern electroanalytical methods, potenciometry with ion selective electrode, elimination and stripping voltammetry. B) UV/VIS spectrophotometry, atomic absorption spectrometry (AAS), atomic emission spectrometry with inductively coupled plasma (ICP-OES), laser spectrometry (LA-ICP-OES, LIBS). C) Isotachophoresis, high performance liquid chromatography (HPLC), capillary chromatography on monolithic stationary phases, capillary zone electrophoresis, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS).
Learning outcomes
Obtaining theoretical knowledge and practical skills in selected analytical methods. Students get to know with hi-tech instrumentation used for research and development of advanced analytical techniques.
Syllabus
  • A) Introduction to course: electroanalytical methods, spectroscopy, separation methods. B) ELECTROANALYTICAL METHODS 1. Nitrate determination by ion-selective electrode in the presence of interfering ion. Nikolski equation. Determination of selectivity coefficient by constant interferent concentration method. 2. Elimination Voltammetry with Linear Scan (EVLS), potential-close signals separation (monitoring of reduction and oxidation on different electrodes). C) SPECTRAL METHODS 3. UV / VIS molecular spectroscopy. Multicomponent spectroscopic and kinetic analysis. 4.Atomic absorption spectrometry with electrothermal atomization 5. ICP OES/MS spectrometry. Methods for decomposition of samples: cryogenic grinding, microwave decomposition. Solution analysis: ICP OES and ICP MS spectrometry. 6. Analytical methods based on laser ablation (LA ICP MS). Laser-induced breakdown spectroscopy (LIBS): surface mapping. D) SEPARATION METHODS 7. Determination of anions by ITP 8. Liquid chromatography (HPLC) - Determination of inosine, adenosine and 2'-deoxy-forms in the model mixture - optimization and validation of the method. 9. Polyacrylamide gel electrophoresis (SDS-PAGE). Separation of proteins in a discontinuous GE system. Subsequent visualization by CBB R 250 and silver. Identification of model proteins in a mixture. 10. CE-LIF, capillary zone electrophoresis with laser-induced fluorescence detection. Optimization of experimental setup. Determination of the detection limit for Rhodamine 6G. Separation of rhodamine dyes. 11. Matrix Assisted Laser Desorption Ionization Mass Spectrometry (MALDI MS). Selected applications of MALDI MS: calibration, determination of molecular weight, peptide mapping, identification of unknown protein.
