- Course objectives
- Bouguert-Lamber-Beer law, absorption spectra, instrumentation, errors, calibration, optimization difference spectrometry, derivative and two-wavelength spectrometry, wavelength modulation spectrometry, extraction spectrometry, sensitivity and limits, complexation in solutions, spectrophotometric reagents, thermal lensing and intracavity laser spectrometry.
- Syllabus
- 1. Origin of discoloration of compounds, absorption spectra, influence of chemical and electronic structure of compounds and ions, isosbestic point. 2. Bouguert-Lambert-Beer's law, apparent and real deviations from its validity. Influence of monochromatization, light scatter, fluorescence, chemical equilibria, refractive index. 3. Instrumentation, spectrophotometer UV/Vis, radiation sources, dispersion modules, detectors. 4. Errors in spectrophotometry, random errors, systematic errors, instrument errors, errors following from chemical equilibria, calibration of spectrophotometer. 5. Study of spectrophotometric characteristics of inorganic systems in visible region, solvated ions, complexes with chromogenic reagents. 6. Study of complex forming equilibria, method of continual variations, method of ratio of tangents, method of molar ratio, absorbance-pH dependences. 7. Interferences, threshold concentration of interfering constituent, positive and negative error, constant and proportional interference, competition equilibria, overlaps of spectral absorption bands, masking agents. 8. Spectrophotometry and organic analytical reagents, selectivity, sensitivity. 9. Application of spectrophotometry UV/Vis in inorganic elemental analysis, examples of employed methods. 10. Extraction spectrophotometry. 11. Spectrophotometry and kinetic methods, differential technique, method of tangents, method of fixed time, method of fixed concentration of reaction product, integral technique, catalytic reactions, enzyme catalyzed reactions. 12. Multicomponent analysis 13. Photoacoustic spectrometry. 14. Statistical evaluation of results.
