Course objectives
This course is for students to gain the basic knowledge about the preparation of proteins in sufficient quantity and quality for crystallization, NMR analysis and others biochemical measurements. At the end of this course the student will be able to: understand the different approaches in the cloning of various expression vectors using affinity tags or without tags, and subsequent purification of the recombinant protein with affinity chromatography or classically; to remove the fusion protein (anchor); to hold the protein in soluble state; to know various methods for protein detection during the purification procedures.
Syllabus
  • 1. The molecular principles for understanding proteins. 2. Calculation in the molecular biosciences 3. The important properties of proteins and how to explore them. 4. DNA cloning. 5. Protein expression. 6. Cell disruption. 7. Fusion proteins and affinity purification. 8. Protein purification chromatography. 9. Protein quality. 10 Analytical methods used to study proteins. 11.
