- Course objectives
- Course contains preparation and cultivation of bacterial viruses; Preparation of phage lysate; Assessing of the phage titre ; One step growth curve of the bacteriophage; Assessing of the burst size of phages; Mutation index; Induction of the phage using UV-light; Transduction in Staphylococcus aureus; Isolation of the temperature sensitive mutants of bacteriophages. After graduation this course, the student should be able to competently work with the viruses originating from the bacteria. After graduation this course, the student should be able to competently analyse the bacteriophages and their genetic material.
- Syllabus
- 1. One step growth curve of polyvalent phage 812 2. Preparing of ts mutant sensitive to high temperature. 3. Estimation of frequency of mutation in phages 4. Kinetics of adsorption, construction of adsorption curve 5. Induction of prophage, delysogenization in the host cells of S. aureus 6. Calculation the force of gene binding, estimation of distance between genes 7. Construstion of genomic map of phages using two- and three factor crossing 8. Estimation of burst-size of phage from single cells of S. aureus or E. coli using Poissons distribution.
