Course objectives
At the end of the course, students should be able to:
- utilize theory of analytical separation for characterisation and understanding of methods based on chromatographic principles;
- understand and clarify principles of instrumentation and functional layout of thin layer chromatography, methods based on principles of chromatography and analytical electromigration, inversed chromatography and hiphenated separation and related analytical technics;
- asses the possibility of combination of chromatographic and other separation and analytical technics for increasing of identification efficiency and improvement of quantification limits of developed analytical methods;
- aplly theory of chromatography to characterisation of analytical phase systems, surfaces of solid materials and bonded phases and to interaction of analytes with components of phase systems;
Syllabus (in Czech)
  • 1. Tenkovrstvá chromatografie. Popis experientálního uspořádání. Charakteristické rysy TLC. Fázové systémy TLC. Postupy, kroky planární chromatografie. Vyvíjení chromatogramu, Techniky řízené kapilárními salami, techniky s nuceným tokem mobilní faze. Gradientová eluce v TLC. 2. Instrumentace TLC. Konstrukce a příprava tenké vrstvy, materiály pro stacionární fáze. Vysoce účinná TLC. Dávkování vzorků, Eluční techniky, volba mobilní faze, klasifikace složek mobilní faze, modely Snyder a Nyiredy,. 3. Detekce v TLC. Přímé moetody, derivatizační postupy. Dokumentace. Vyhodnocení TLC 4. Vícerozměrná TLC. Oblasti použití TLC. 5. Hybridní metody, elektrochromatografie, micelární elektrokinetická chromatografie, mikroemulsní elektrokinetická chromatografie, kapilární chirální elektroforéza, gelová elektroforéza. Experimentální podmínky. 6. Pohyb iontu v elektrickém poli, základní rovnice, pojmy a parametry. Elektroosmotický tok. Kapilární elektroforéza (CE), Izotachoforéza, izoelektrická fokusace. Přechodová isotachoforéza. 7. Instrumentace a postupy hybridních technik. Ddávkování vzorků, Detekce 8. Inversní chromatografie. Definice, zařazení v chromatografických metodách 9. Oblasti použití inverzní chroatografie. Výstupy. Studium adsorpčních vlastností pevných materialů, měření adsorpční izotermy –Glueckauf, Gregg/Stock. Adsorpční teplo. 10. Oblasti použití inverzní chroatografie. Mezimolekulové interakce. Porozita chromatografického lože. Studium vlastností imobilizovaných polymer. Humínové kyseliny, biopolymery. Aktivní centra na povrchu pevné faze. Katalytická aktivita. Fázové přeměny. Elekrokinetické studie. 11. Kombinované techniky v separační analýze. Vícerozměrné separace. Spojení LC-GC. On-line extrakce a předseparace, SPE, SPME, SFE, ASE. Spektrální metody pro on-line detekci a identifikaci, GC-MS, LC-MS, GC(LC)-ICP-MS, LC-NMR, LC-FTIR. 12. Trendy v chromatografii. Vícerozměrová chromatografie. Vysoce rychlá chromatografie. Mikroseparace.

Teaching methods
Education is performed as lectures with Powerpoint prezentation. Understanding of mechanisms and consequences is emphasized. Students are frequently asked questions to think about actual topic. They are encouraged to ask questions and be in interaction with the lecturer.
Assessment methods
Attendance of the lectures is not mandatory but strongly recommended for fluent understanding of the educated subjects. Final assesment (at the end of semester) is by oral examination. Tree related topics are the content of the discussion during oral examination.
