Flagella motion – separated drop
1. Drop of culture suspension on microsopic slide Do not cover by cover
glass and use a microscope to observe
2. Focus carefully on the upper part of the drop
– use 20x and maximum 40x objective
Staining of flagella
1. Drop of culture suspension on microsopic slide mix with dye of staining of
flagella and cover with cover glass
2. Observe preparation at magnigication 1000x with immersion oil
Observing of motion on agar for movement testing
1. Innoculate agar by one stab in the middle of agar
• to grow only 1 colony
2. Innoculate Petri dish and also tubes
What to observe ?
o Flagella motion (1)
o Staining of flagella (1)
o Observing motion on agar:
• 2x Petri dish
• 2x tube
Bacilli:
Bacillus cereus
Cocci:
Micrococcus luteus
Rods:
Pseudomonas fluorescens
Escherichia coli
