- Main objective of the course is to understand the basic approaches of the proteomics, i.e. structural and functional study of proteins using recombinant proteins technology, protein separation using different techniques, predominantly two-dimensional electrophoresis and liquid chromatography and basic techniques of protein mass spectrometry. Students will be able to apply those approaches during solution of recent problems of both basic and applied research.

- Course objectives
- In the course the students will be educated in the basics of the experimental approaches recently used in the studies of the function of the sum of all proteins of the respective organism (proteome), but on the other hand also to the study of the features of individual proteins. Students will practice the methods of protein expression and purification in heterologous expression systems (E. coli), ranging from the recombinant DNA isolation and characterization, through the protein isolation, purification and purity analysis using SDS-PAGE and image analysis to the structural and functional analysis of purified proteins. Further there will be performed total protein isolation from the model organism Arabidopsis thaliana and students will be educated in the basic approaches of the proteome analysis using 2D electrophoresis and its evaluation using image analysis. It will be followed by the practical demonstration of the basics of the protein identification using MS spectrometry and the basics of the work with protein databases. All the above mentioned methods and methodological approaches are used in the Laboratory of Plant Molecular Physiology used for the solution of real biological and biochemical problems by the staff and students of the laboratory that will participate on the teaching of the course.

- Pro úspěšné absolvování kurzu a udělení zápočtu bude požadováno splnění všech studijních povinností a to nejpozději v POSLEDNÍ DEN konání daného běhu (tedy vždy v pátek). Jedná se především o absolvování několika písemných testů a odevzdání VŠECH požadovaných protokolů.

- Main objective of the course is to understand the basic approaches of the proteomics, i.e. structural and functional study of proteins using recombinant proteins technology, protein separation using different techniques, predominantly two-dimensional electrophoresis and liquid chromatography and basic techniques of protein mass spectrometry. Students will be able to apply those approaches during solution of recent problems of both basic and applied research.

- Two-dimensional electrophoresis of proteins
- Mass spectrometry of proteins (MALDI-TOF, LC-MS/MS, sample preparation and analysis, protein identification)
- Preparation, isolation and sequencing of recombinant DNA
- Expression of recombinant DNA in E.coli
- Single and multistep purification of recombinant proteins
- Purity determination and analysis of recombinant proteins
- Western bloting
- Literature
- Teaching methods
- Practical solution of problems demonstrating utilizing of basic instruments of recent proteomics.
- Assessment methods
- The course will and up by colloquium. The prerequisite for successful completion of the course is timely submission of required protocols and participation in the final interview.
