- Course objectives
- At the end of this course students should be able to understand and to explain:
the basic terms in toxicology and chemical safety;
adverse local, acute and chronic systemic and specific late effects, physical hazards and ecotoxicity of substances and mixtures;
the methods and guidelines of testing, assessment, classification and labelling of hazardous substances and mixtures;
the factors determining the adverse effects of substances on human health, the interaction of substances with organism - exposure, absorption, distribution, biotransformation and elimination of xenobiotics, molecular, cellular and organ levels of interaction;
Czech and EU legislation on substances and mixtures, public health and occupational safety and health;
occupational exposure limits, maximum permissible concentrations of substances in water, maximum residue levels in foods;
the principles of the risk analysis and the protection of workers from the risk related to exposure to chemicals, emergency measures in cases of accidental substance exposure;
hazardous properties of important inorganic and organic mixtures;
sources of information on hazardous properties of substances and mixtures. - Syllabus
- 1. Introduction, aims and programme of course. The basic terms: toxicology, chemical safety, harmful chemicals, xenobiotics, exposure, dose, effect, latent period, response, hazard, risk, classification, hazardous substances and mixtures, hazard pictograms, signal words, risk and safety phrases, hazard and precautionary statements. Local effects, the testing and evaluation of acute irritation and corrosion, the substances and mixtures corrosive and irritant to skin and causing serious eye damage or eye irritation.
- 2. Systemic effects, acute and chronic intoxications, the testing and evaluation of acute, subacute, subchronic and chronic toxicity, LD50, LC50, ATE, NOAEL, LOAEL, the substances and mixtures very toxic, toxic and harmful, substances and mixtures with acute toxicity, specific target organ toxicity after a single and repeated exposure and aspiration toxicity. Sensitisation, allergy, the testing and evaluation of sensitisation, respiratory and skin sensitisers.
- 3. Specific late effects. Mutagens, gene mutations, structural and numerical chromosome aberrations, genetic toxicology, bacterial reverse mutation tests, mammalian bone marrow chromosome aberration test, DNA damage and repair tests, epidemiological studies, the evaluation of mutagenicity of chemicals, germ cell mutagenic substances and mixtures.
- 4. Carcinogenicity, carcinogenesis, carcinogenicity studies, epidemiological studies, the evaluation of carcinogenicity, carcinogenic substances and mixtures.
- 5. Reproductive and developmental toxicity, embryotoxicity, teratogenicity, testing of reproductive toxicity and teratogenicity, epidemiological studies, evaluation of reproductive toxicity, substances and mixtures toxic for reproduction.
- 6. Harmful physical properties, their testing and evaluation, explosive, oxidising, extremely flammable, highly flammable and flammable substances and mixtures.
- 7. Ecotoxicology, the testing and evaluation of ecotoxicity and other properties of substances and mixtures which are harmful for the environment.
- 8. The factors determining the adverse effects of chemicals on organism: substance, organism, dose, the others.
- 9. The interaction of substances with organism. Routes of exposure, absorption, distribution, biotransformation (the reactions of the first and second phase of metabolic conversion) and elimination of xenobiotics, molecular, cellular and organ levels of interaction. Exposure tests, biological threshold limit values, genetic risk monitoring of workers exposed to genotoxic agents.
- 10. The European Union legislation on substances and mixtures - Regulation No 1907/2006 (REACH), Directive 67/548/EEC, Regulation No 1272/2008 (CLP). The Czech legislation on substances and mixtures, the public health protection, occupational safety and health protection, and the prevention of major accidents. Permissible exposure levels and short-term exposure limits of chemicals and dusts in the air in the working zone, permissible concentrations of chemicals and dusts in indoor environment, maximum permissible concentrations of substances in drinking water, maximum residue levels in food.
- 11. The risk assessment and occupational health protection of workpeople, individual protective devices, the first aid measures in cases of accidental chemical exposure.
- 12. Special toxicology of inorganic substances.
- 13. Special toxicology of important organic compounds (aliphatic and aromatic hydrocarbons, halogenated hydrocarbons, alcohols, phenols, ethers, aldehydes, ketones, carboxylic acids and their derivatives, inorganic acid esters, nitro compounds, amines, organometallic compounds).
- 14. The information sources on dangerous properties of substances and mixtures. Safety data sheets, toxicological literature, databases on CD-ROM and on-line, toxicological information centre.
