- Course objectives
- This practical course is conceived as an introduction into methods of molecular biology.
- Syllabus
- 1. Preparation of solutions and stock enzymes used in molecular biology.
- 2. Isolation of plasmid DNA (pUC family, pBluescript). Determination of DNA concentration and purity.
- 3. Restriction analysis of nucleic acids, agarose gel electrophoresis.
- 4. Restriction mapping, isolation of DNA from agarose gels.
- 5. DNA cloning in basic types of vectors, DNA transfer to prokaryotic cells, transformation, selection of clones.
- 6. Centrifugation techniques, preparation of sacharose density gradients.
- 7. Isolation of bacterial genomic DNA for DNA fingerprinting.
- 8. Polymerase chain reaction and their modifications (PCR-RFLP, AP-PCR).

- Teaching methods
- The practical course is taught in seven 6-hour blocks in a molecular biology laboratory. Students perform the methods according to examined protocols.
- Assessment methods
- The course is completed after work out an elaborate from the performed tasks and pass a writen test that contained 20 practically oriented questions.
