Course objectives
Students will be able consider the sutiable technique for observing microscopic and macroscopic characteristics of bacteria and their morphological diversity (including the cell morphology of particular bacterial species depending on cultivating media) and observing structures of bacterial cells (endospores, inclusions, flagellum). Student will be able consider the applicability of particular methods for clinical, environmental and biotechnological practice. Student will be able of preparing of variable stained and native microscopic slides, will be able operate variable microscopical techniques (phase and Nomarsky contrast, bright field microscopy) and handle with images acquired by CCD camera, photographic camera and analyse them (image acquisition - LUCIA G).
Syllabus
  • Morphology of bacterial and archaeal cells and colonies, basic staining methods.
  • Techniques of work with light microscope, care, removing of simple problems.
  • Microphotography, digital photography, the use of video-cameras in image acquisition in microbiology, computer image analysis.
  • Determination of the size of bacterial cells and their morphological structures (the use of ocular micrometer and software for image analysis).
  • Gram staining. Acid-fast bacteria, Ziehl-Neelsen staining, mycobacteria. Giemsa staining.
