- Course objectives
- The participant will master advanced techniques of plant cytogenetics (microscopic preps, advanced fluorescence microscopy, chromosome painting, genomic in situ hybridization, image processing).
- Learning outcomes
- Student will be able to:
- prepare micrscopic preparations;
- label isolated DNA by haptens and fluorochromes;
- prepare DNA probes;
- carry out fluorescence in situ hybridization (FISH) of the labeled DNA probes to chromosomes and cell nuclei;
- analyze preparations using fluorescence microscopy;
- photograph chromosomes and cell nuclei;
- analyze and process the acquired photographs. - Syllabus
- Introduction to evolutionary and comparative plant cytogenetics. Principles of light and fluorescence microscopy. Preparation of mitotic and meiotic chromosomes from roo tip meristems and anthers, microscopic analysis of chromosome preparations. Direct and indirect labelling of DNA probes by nick translation. Fluorescence in situ hybridization (FISH), genomic in situ hybridization (GISH), comparative chromosome painting (CCP). Immunodetection indirectly labelled DNA probes. Fluorescence microscopy of chromosome preps after FISH, GISH a CCP. Processing of microscopy photographs (Adobe Photoshop), interpretations.
