Research experience
  • Chemistry instrumentation development
    In cooperation with my supervisor and technician in our department I constructed and tested 532 nm laser induced fluorescence detection system for Capillary Electrophoresis. I was responsible to measurement, improvement and service of CELIF and CEUV instrument for 5 years. I gained practical knowledge about setting optics, photomultiplier tubes (PMT) applications, solid state lasers application, fluorescence measurements and data aquisition throughout I/O digitizing PC card.
    During my Ph.D. study I constructed and tested subatmospheric pressure deposition device for coupling CE separation with MALDI MS detection. I gained practical knowledge about microfluidic technology, coupling CE to MS, crystallizing process and vacuum technology.
    I learned application programming for PMT data aquisition and driving stepper motors in NI LabVIEW 6.1 software.
  • Proteomic Analysis
    Aim of my Ph.D. thesis is an alternative proteomics analysis of bacteriophage 812 proteins utilizing developed platform mentioned above (protein fraction>CE-deposition>on-target LIF identification>on-target digestion>MS). The obtained proteomic data are evaluated to genome database, gained proteome map is compared to classic proteomic approach (protein fraction>PAGE>digestion in solution>MS). The main advantages of the alternative approach are higher sensitivity, very low consumption of a sample and complementary information output to the classic approach.
  • Methodology
    Column Separation: I gained experience with CE, ITP and HPLC separation of rhodamine based dyes, aminoacids, peptides and proteins, nucleotides and fluorescent derivatized biomolecules containing amino groups.
    Fluorescence: Measuring and detection of fluorescence in-solution or in-solid state after excitation with Xe lamp or 532/266 nm solid state lasers. I studied effect of organic solvents/water electrolytes to fluorescence in solution and effect of additives in case of solid state fluorescence.
    Mass Spectrometry: I have 5 years experience with MALDITOF MS measurements of peptides and proteins, rhodamine based dyes derivatized peptides & proteins and digested proteins. Automation of mass spectrometric measurement & data processing.
