Course objectives
The aim of the theoretical part of the preparation is to provide detailed information on methodologies using the latest knowledge of the field with respect to the current state of routine application of new techniques and technologies. Fundamentals of indication and interpretation of laboratory methods in relation to clinical units and diagnoses. In the practical part of the training, gain practical skills of such a magnitude that allow the graduate to be easily assigned to clinical laboratory teams;
Learning outcomes
Upon completion of the course the student will be able to: actively assess the correctness of indication and interpretation of biochemical laboratory examination; to reliably detect and respond to instances of interference; communicate appropriately with clinicians; to respond appropriately to outlier findings; to have an overview of the used analytical principles and to know their limits; to follow standard analytical, technical and logistical operations.
Syllabus
  • Laboratory sample reception and identification of biological material - paper and electronic requests, bar codes, readers and bar code printers
  • Laboratory Information System - individual LIS modules, on-line and off-line communication methods, pipetting sheets, print assemblies and outputs, system of controls, supervision, reports of performance, monitoring of the economy
  • Preparation of analytical samples - sampling vessels, centrifugation, pipetting, sorting
  • Work with automatic biochemical analyzers - chemical parameters of methods, linear and nonlinear calibration, preparation of reagents, reaction tracking, lag-phase, linearity, substrate depletion
  • Installation of end-point method, monochromatic and dichromatic measurements, calibration
  • Installation of a kinetic method for enzymatic catalytic activity determination
  • Chemical and morphological examination of urine - manual and automated operation
  • Measurement of serum and urine osmolality, solutions of ions, glucose and urea - osmolality calculations
  • Measurement of acidobasic metabolism and blood gases parameters (pH, pCO2, pO2), effect of air bubbles on the measured parameters
  • Electrophoresis of proteins on agarose, densitometry, immunofixation
  • HPLC - sample preparation, vanillylmandelic acid, HIOK
  • Atomic Absorption Spectrophotometry - Determination of Mg, Ca, Zn Flame technique, Cu determination by electrothermal atomization
  • Morphological examination of cerebrospinal fluid - quantitative cytology in the counting cell, preparation of cytospin slide
  • Biochemical examination of cerebrospinal fluid, spectrophotometric curve
  • System of Internal and External Quality Control - Westgard Rules, Implementation, Evaluation
  • Comparison of two methods, correlation, Passing-Bablock
  • Documents of good laboratory practice and quality management system according to ISO standards
  • Practical exercises are organized in groups of 6 students, each working individually with the writing and processing of measured data in the laboratory protocol. The exercises take place in the laboratory of the Department of Laboratory Methods (automatic biochemical analyzers: MODULAR ISE-P on-line connected to LIS-Infolab including barcode printers, HITACHI 911, Cobas Mira plus, photometer with a heated cell- FLM-3 Radiometer; Atomic Absorption Spectrophotometer-Flame Technique; Osmometer; BioRad Electrophoresis; pH Meter; Mettler Analytical Balance and Metroller Transfer; Thin Chromatography TOXILAB-Merck; Cytofuge; 10pcs of Microscope for Individual Work; students at the same time; HD-camera microscope and projection equipment; Some practical exercises take place in the laboratories of OKB FN Brno (Immunochemical analyzers, ELISA, RIA, automated urine analysis, HPLC, AAS-electrothermal atomization, acid-base analyzers + blood gases.

individual of solving problems from the analytical part of the subject using analyzers, instruments and laboratory equipment in the teaching laboratory and in the routine operation of the clinical laboratory; Writing a protocol with measured values, evaluation and conclusion; Continuous monitoring of assigned tasks, possible corrections and evaluation by the lecturer;

Credits are obtained after active participation of the practices, finished protocols and examination of gained knowledge and practical skills
