Syllabus
  • The cell cycle (phases, control points, role of cyclins in regulation, methodical approach for analysis of the cell cycle, genetical studies of the cell cycle in yeast). Cell signalling (sorting of cellular signals, hormonal signalling, role of intracellular and cell surface signal receptors, intracellular signal pathways, role of G proteins, cAMP, Calcium ionts and protein kinases in cellular signalling). Nervous system (neuron, synapsis, action potential, molecular properties of voltage-gated ion channel proteins, myelin, neurotransmitters, structure of acetylcholin receptors). Cell-to-cell and cell-to-extracellular matrix interactions in multicellular organisms (structure and function of different types of matrix, molecular structure of collagen, hyaluronic acid and proteoglykans, laminin and fibronectin, cadherins, neural cell adhesion molecules). Molecular immunology (differentiation of hematopoietic cells, role of growth factor in hematopoiesis, lymhokines and monokines, function of interferons and interleukins and their practical applications, tumor necrosis factor, monoclonal antibodies: preparation and usage, antigen processing, structure and function of MHC class I and II molecules). Molecular basis of carcinogenesis (properties of malignant cells, role of oncogenes, tumor suppressors, programmed cell death regulators in carcinogenesis, molecular mechanisms of carcinogenesis, cancer and the cell cycle, protooncogenes and their products, cooperation of oncogenes in malignant transformation, tumor immunology, immunodiagnostics of tumor cells, immunotherapy). Muscle structure and function (molecular basis of nerve - muscle cell connection, structure of thick and thin filaments, role of actin, myosin and troponin in muscle contraction, role of transcription regulation in differentiation of muscle cells, differentiation of muscle cells in vitro - role of MyoD1, myogenin, MRF4 and Myf5, helix-loop-helix structure of MyoD protein). Yeast - useful single cell model for investigation of eukaryotic cell cycle, differentiation, chromosome structure and replication (cell type determination in yeast - example of differentiation in single cell eukaryots, yeast life cycle, molecular mechanism of mating type conversion, role of MAT, HML, HMR and HO locuses, transcriptional regulators and cell type determination - gal genes expression in yeast, structure of GAL4 protein, upstream activating sequences, structure of artifitial yeast chromosomes, structure and function of centromere, telomere and ARS). Tissue cultures (principles of cell cultivation, primary and secondary cultures, embryonic stem cells, differentiated cells in culture, viral infection of cells in culture, transfer of DNA into the cells in culture, transient and permanent transfection).
