- Course objectives
- Basic aspect of light microscopy with a view to the zoological methods - dark field, phase contrast, differential interference contrast, fluorescence microscopy,measurement methods, drawing, photomicrography). Basic zoological techniques - native and permanent slides, fixation, staining, mounting media, embedding and cutting.
- Syllabus
- 1. Basic aspects of light microscopy, types of microscopy,the compound microscope and Köhler illumination,lens nomenclature 2. Measurements methods, calibration, ocular micrometer, stage micrometer, digital analysis, drawing 3. Contrast methods, dark-field, phase-contrast, differential interference contrast (DIC) 4. Photomicrography, digital photomicrography 5. Native slides, vital staining, Neutral red, Congo red, Methylen green 6. Fixation,properties of some common chemical fixatives 7. Aqueous mounting media, glycerol,glycerol-gelatine, Liquido-Faure, PVA 8. Canada balsam, dehydration, clearing 9. Stains and staining,hematoxylin staining, carmine staining 10.Histological methods, clearing, infiltration, embedding for paraplast method 11.Sectioning, mounting and staining 12.Fluorescence microscopy, fluorescence
