- Basic knowledge of biochemistry is prerequisited (C3181 Biochemistry I, C4182 Biochemistry II). C7895 Mass Spectrometry of biomolecules is a recommended lecture

- Course objectives
- Subject summarises the basic knowledge of the analytical approaches in analysis of biomacromolecules snd suprabiomolecular structures and also important theoretical background for their understanding.
- Syllabus
- - analytical approaches in v bioanalytics demands; high throuput, shotgun analysis, standardisation; affinity methods principles, conduct and use (separation, interaction studies), IMAC, APAGE
- - immunoanalytic methods principles; immunoanalysis precipitation, agglutination, immunoessay, methods: FIA, LIA, RIA, EIA, ELISA, IAC, IACE, SELDI
- - gel electrophoresis principles, conduct (1D, 2D); blotting
- - genomics (analysis of DNA) fundaments of genetics (structura, function of DNA), analytical approaches: PCR, restriction enzymes; sequencing; DNA methylation and its analysis
- - proteomics (analysis of proteins and their complexes) fundamentals of proteomics (structure of proteins and protein complexes); expression proteomics methodology, methods (2D HPLC, 2D PAGE, MS: bottom-up, top-down),
- - functional proteomics - methodology, methods (TAP-tagging, Two-Hybrid, SEAM strategy)
- - bioinformatics in MS-assisted proteomics identification of protein in organisms with and without sequenced genoms (MS BLAST, MultiTag)
- - quantitative analysis of protein complexes internal standard method; mass tagging (ICAT, ICMT, TMT, MCAT, GIST, SILAC, SIL), absolute quantification of proteins and peptides by MS
- - structural proteomics methodology (chemical cross-linking), methods (FTICR MS)
- - lipidomics (analysis of lipids and cellular membranes) fundamentals in lipidomics (structure and function of cell. membranes and their components), analytical approaches: lipid extraction, TLC, GC, LC, SFC, GPC, MS (PIS, HGS, FAS)
