Molecular structure a function of procaryotic and eucaryotic genome. Cultiovation of bacteria, estimation of titre, CFU.

Course objectives
The subject is a part of basic course Molecular biology for students of discipline Molecular and cellular diagnostic. It comprises the tasks that offer acquiring of basic methods in molecular biology of bacteria and their viruses.
Syllabus
  • 1. Makrorestrikční analýza genomové DNA pomocí pulzní-gelové elektroforézy. 2. Neradioaktivní značení sond digoxigeninem. 3. Southernův přenos a selektivní hybridizace bakteriální DNA se specifickými sondami. 4. PCR detekce specifických genů u bakterií. 5. Izolace proteinu z zivocisnych bunek, elektroforeza SDS-PAGE, westernovy prenos, protilatkova detekce specifickeho proteinu. 6. Stanovení počtu živočišných buněk v populaci s použitím hemocytometru a mikroskopie. 7.-12. Specializované úlohy z molekulární diagnostiky prováděné na externích odborných pracovištích.
