Course objectives
This course will introduce principles and applications of modern methodologies of analytical cytometry such is flow cytometry, sorting, digital microscopy and microarrays. Analysis of data and their interpretation and presentation will be explained in details. Part of the course is also practical demonstration of selected equipments and methods of data analysis. We will put stress on possibilities and methods for practical applications in medicine and biology.
Syllabus
  • * Flow cytometry general introduction - basic principles and history (Karel Souček)
  • * Principles of flow cytometry and sorting (Karel Souček) -fluorescence -excitation sources, optical systems and detection of fluorescence -signal processing and compensation -data visualization, analysis and presentation
  • * Biological application of flow cytometry (Karel Souček) -cell biology (nucleic acid analysis, cell function analysis, flow cytometric methods in cell death analysis, principles and application of multiparametric methods) -cytogenetics -hydrobiology
  • * Application of flow cytometry in clinical immunology and hematology (Lukáš Kubala)
  • * Principles of digital microscopy (Eva Bártová) -fluorescent microscopy, confocal microscopy, temporally resolved -digital microscopy, time/frequency resolved digital microscopy
  • * Clinical and biological applications of digital microscopy (Eva Bártová) -chromosomal and gene analysis -metabolism and cellular compartments analysis (pH, ions) -FRET and FRAP applications
  • * Microarray analysis principles (Pavla Gajdušková) -DNA, RNA arrays -expression, gene and mutational arrays
  • * Biological and clinical application of microarrays and data analysis (Pavla Gajdušková) -practical examples of application in diagnostic and basic research
Assessment methods (in Czech)
Kurz bude postaven zejména na přednáškách a diskuzích k nim. V rámci přednášek budou objasněny principy a způsoby základní analýzy dat k jednotlivým metodologiím. Studenti formou instruktáže seznámeni s vybavením pro průtokovou cytometrii a digitální mikroskopii které je v provozu na Biofyzikálním ústavu v Brně. Zkouška Každý student bude muset prezentovat krátký seminář jehož téma bude konzultováno s přednášejícím a bude se týkat zaměření kurzu. Na základě této prezentace bude udělen zápočet a hodnocení vlastního semináře se bude také z 30-ti % odrážet v celkové známce. Kurz bude zakončen zkouškou ve formě testu shrnujícího látku za celý semestr. Výsledek testu bude tvořit 70% celkového hodnocení.
