- Course objectives
- The main objective of the course is to provide the students with the ability to
- list basic techniques of optical microscopy and explain the physical principles of these techniques
- apply the correct technique for a particular sample and examine this choice in terms of the obtained image contrast,
all for the micro structured samples in biology, medicine, mineralogy and metallurgy. - Syllabus
- 1. Optical imaging, thin lense, optical system, main planes, focal planes 2. Construction beams, real imaging beams, entrance diaphragm, angular aperture 3. Limitting resolution, depth of field 4. Beam splitters, reflectivity and transmitivity, total relfection, beam divergence 5. Spectral composition of light, light absorption, light detectors, human eye 6. Optical microscope, coherent and noncoherent illumination of sample, field lense 7. Sample properties, absorption, double refraction, scattering, reflectance 8. Method of dark field in transmition and reflection 9. Double beam interference, time and spatial coherence, visibility 10. Imaging contrast in transmission and reflection in interference microscope. 11. Linear polarization of light, polarizer, transmission of light in double reraction materials 12. Polarization microscope, molecular structure of samples and double refraction. 13. Principles of scanning computer optical microscopy methods 14. Imaging od 3D samples, contrast by scanning methods, optical tomography

- Teaching methods
- Integral part of this course is the excersize during which the topics from the lectures are worked through. The content of the excersize is formed by examples of the basic ray construction for the selected imaging aparatuses and/or methods.
- Assessment methods
- For successful completition of the course, passing the final written test is necessary. Unless stated otherwise, 80% attendance at the exercises is mandatory.
