- Course objectives
- At the end of the course, the students should be well informed about the selected methods of collection of haematophagous arthropods, they should be able to work safely in laboratories with zoonotic microorganisms, to prepare cell cultures, and to use basic molecular methods apllicable for detection of microorganisms in haematophagous arthropods (PCR, RT-PCR, real-time PCR).
- Syllabus
- 1. Demonstration of collection techniques for selected haematophagous arthropods (especially ixodid ticks and mosquitoes) in the field; determination of arthropods in the laboratory. 2. Examination of microscope slides and colour figures of zoonotic pathogens. 3. Guidelines for safe work in the laboratory handling zoonotic microorganisms - safety rules in infectious environment (the work in biohazard and laminar boxes, material decontamination). 4. Serological methods – detection of antibodies to selected vertebrate pathogenic viruses (PRNT, ELISA): demonstration of the tests. 5. Basics of the work with cell cultures: practical steps - preparation of the cultures, their infection with a virus, and inspection for cytopathic effect. 6. Molecular techniques used for the detection of microorganisms in haematophagous arthropods (PCR, RT-PCR, real-time PCR), demonstrations - isolation of DNA and RNA; PCR; gel electrophoresis, photodocumentation of the gel, purification of PCR products).
