- Course objectives
- At the end of the course, students should be able to:
- define fundamentals of mass spectrometry and describe it in the context with other spectral analytical methods;
- understand and clarify principles of instrumentation and functional layout of current ionization techniques, mass analyzers and detecting elements;
- explain the mechanisms of ion fragmentation and dissociation using current ionization techniques and fragmentation processes;
- asses and interpret the mass spectra of common organic and anorganic compounds obtained by most frequently used ionization techniques;
- evaluate significance of connection of mass spectrometry with various analytical techniques, especially with separation techniques GC/MS, HPLC/MS, CE/MS, ICP/MS;
- take advantage of mass spectroscopy in quantitative and qualitative analyses of various types of samples; - Syllabus
- I. History, principles of MS, fundamental terms.
- II. Instrumentation. Sample introduction, vacuum system. Sample ionization, methods of ionization of volatiles / nonvolatiles compounds, soft and hard ionization techniques. Ion analysis, resolution, magnetic sector, electrostatic analysator, HRMS. TOF analysator and MALDI-TOF equipment. Ion cyclotron resonance. Linear quadrupole, ion trap. Tandem MS. Collisional activation. Ion detection. Spectrometer tuning.
- III. Fragmentation. Metastable ions, nuclide ions, fundamental fragmentation mechanisms.
- IV. Mass spectra and their use. Quantification in MS.
- V. Hyphenated techniques. Hyphenation with separation techniques GC/MS, HPLC/MS, CE/MS. Data acquisition and analysis. ICP/MS technique.

- Teaching methods
- Education is performed as lectures with Powerpoint presentation. Understanding of mechanisms and consequences is emphasized. Students are frequently asked questions to think about actual topic. They are encouraged to ask questions and be in interaction with the lecturer.
- Assessment methods
- Attendance of the lectures is not mandatory but strongly recommended for fluent understanding of the educated subjects. Final assessment (at the end of semester) is by oral examination. Tree related topics are the content of the discussion during oral examination.
