- Course objectives
- Introduction to molecular biology methods performed in the respective practice. After completing the course, students should know the principles of methods of isolation and analysis of genetic material such as plasmid and chromosomal DNA or cloning.
- Learning outcomes
- The student will be able to describe the principles of molecular biological methods: plasmid DNA isolation; spectrophotometry; refractometry; ultracentrifugation; restriction endonuclease cleavage, electrophoretic separation methods; PCR; cloning including ligation and transformation; approaches to selection of transformants.
- Syllabus
- 1. Preparation of solutions and stock enzymes used in molecular biology. 2. Isolation of plasmid DNA and Determination of DNA concentration and purity. 3. Restriction analysis of nucleic acids, agarose gel electrophoresis. 4. Restriction mapping, isolation of DNA from agarose gels. 5. DNA cloning in basic types of vectors, DNA transfer to prokaryotic cells, transformation, selection of clones. 6. Centrifugation techniques. 7. Isolation of bacterial genomic DNA for DNA fingerprinting. 8. Polymerase chain reaction (PCR) and its modifications (PCR-RFLP, AP-PCR).
