Course objectives
The practical course intended for students in the early stages of their research career, will deal overexpression of proteins in prokaryotic systems with different expression vectors, strategy of protein purification and their characterisation. At the end of this practical course the student will be able to: use the bioinformatics in analysis and planning of a protein expression project; design primers and optimize gene expression; compare in vectors containing different tags and fusion proteins and different strains; optimize the quality of your favorite protein during different purification strategies.
Syllabus
  • 1.Strategy of cloning into expression plasmids. 2. Expression and purification of the human scFv produced in cytoplasm or periplasmic space. 3. Expression and purification of the plant protein (A. pET32 – thioredoxin - chaperon/B. pRSET – His.tag/ C. pET161DEST – Gate way technology – Lumio.tag/D. pET28 – without any tag.)
