- Course objectives
- The main objective of this class is to get an complex overview about human and environmental risk assessment methods and its applications.
- Learning outcomes
- At the end of this course, students should be able to:
- define human and environmental risk assessment methods
- apply prospective and retrospective methods of human and ecological risk assessment
- explain how different exposure parameters can influence final risk predictions
- define toxicokinetics of the main exposure routes (oral, inhalational, dermal, dietary exposure)
- characterize and quantify significant potential risks
- recognize main exposure factors, which have significant effect in total risk assessment
- interpret acceptable carcinogenic and non-carcinogenic risk level
- solve practical case study with risk quantification from define exposure scenarios. - Syllabus
- Introduction - basic concept definition: hazard, risk, risk assessment and risk management, prospective and restrospective approach.
- Reasons of risk assessment application: local, regional and global effects, risk perception, interpretation of exposure.
- Main risk assessment component: hazard identification, dose - response relationships, exposure analysis, risk characterization, risk management.
- Application of risk assessment method in exposome approach – external and internal exposure assessment, (differences in NOAEL x BMD approach).
- Methods of human health risk quantification: basic toxicokinetic models, carcinogenic and non-carcinogenic risk assessment methods – detail procedure, software applications (PC lecture room)
- Description of exposure parameters for different exposure pathways and scenarios (Sensitivity analysis, Exposure Factor Handbook) – inhalation, dermal and dietary exposures. Probabilistic assessment (Monte Carlo approach).
- Physiologically Based Toxicokinetic Modeling (PBTK) – application in Risk Assessment (for exposure backward reconstruction). The description of ADME (absorption, distribution, metabolism, and excretion), Experimental methods for transport coefficients investigation (dermal, gastrointestinal) and QIVIVE application - Quantitative in vitro – in vivo extrapolation.
- New projects with Human health risk assessment approaches; new database sources.
- Human Biomonitoring and application of its results in human health risk assessment (biomarkers; cohort studies; application of long-term biomonitoring of mother milk in CR).

- Teaching methods
- Education is performed as lectures with Powerpoint prezentation. Understanding of mechanisms and consequences is emphasized. Students are frequently asked questions to think about actual topic. They are encouraged to ask questions and be in interaction with the lecturer.
- Assessment methods
- Attendance of the lectures is not mandatory but very strongly recommended for fluent understanding of the educated subjects. During the lectures, students are asked about subjects of past lecture. Final assesment (at the end of semester) is by written examination. Students must successfully realize practical case study with risk quantification from define exposure scenarios (Individual Case Study).
