- Course objectives
- The aim of the course is to enable students to gain practical knowledge of basic molecular-biological methods used in pharmacogenomics and pharmacogenetics (DNA isolation, PCR method, restriction cleavage, agarose gel electrophoresis, etc.).
- Learning outcomes
- At the end of the course, the student will be able to identify and summarize important features of the main methods used in pharmacogenomics and pharmacogenetics. Will be able to practically perform basic molecular-biological analyses and interpret the obtained results.
- Syllabus
- General overview of topics:
1. Isolation of genomic DNA
2. Characterization of DNA, determination of its concentration and purity by spectrophotometry
3. Nucleic acid electrophoresis in agarose gel
4. Genotype determination by PCR, PCR-REA, multiplex PCR, real-time PCR, allele-specific PCR
5. Consultations, submission of protocols
6. Final test
DNA isolation, determination of CCR5 genotype by PCR
Electrophoresis of CCR5 amplicones, determination of ICAM-1 genotype by PCR-REA
Restriction cleavage of ICAM-1 amplicons, electrophoresis of NOD2/1007fs genotype determination by multiplex PCR
Electrophoresis of amplicons of amplicons multiplex PCR, determination of genotype predisposing to celiac disease by allele-specific PCR
method Electrophoresis of coeliac amplicons, determination of Leiden V allele by real-time PCR

- The final assessment is a written test (the test consists of 10 questions, 50% of correct answers is needed to pass). During the semester, it is required to prepare protocols from exercises.
