Course objectives
At the end of the course students should be able to understand and explain - basic properties of natural substances (aminoacids, carbohydrates, lipids, nucleic acids, proteins, enzymes and coenzymes, their properties and structure, active site), thermodynamics, enzyme kinetics, inhibition, allostery, regulation of enzyme activity.
Learning outcomes
Student will be able to: - Perform basic laboratory operations in the biochemical laboratory - Control common laboratory instruments used in the biochemical laboratory - Mathematically evaluate measured data - Interpret the simple results of his work
Syllabus
  • 01. Work safety rules. Organization of laboratory exercises. Preparation of dilute solution, pipetting, and photometry.
  • 02. Isolation, analysis, and determination of deoxyribonucleic acid (DNA).
  • 03. Qualitative and quantitative determination of amino acids.
  • 04. Quantitative determination of proteins.
  • 05. Qualitative and quantitative determination of carbohydrates.
  • 06. Lipids and their analysis.
  • 07. Sucrase - immobilization, substrate specificity, activity determination.
  • 08. pH optimum of enzyme reaction.
  • 09. Enzyme kinetics.
  • 10. Analytical use of enzymes.
  • 11. Biological electron transfer chains.

The first exercise is devoted to learning about safety in     biochemical laboratory. Students also write the entrance test, whose successful completion is a prerequisite for participation in the exercise. Before each next exercise is a written test verifies the student's preparation for the job. In all tests, including entrance test, the student must correctly answer at least 60% of the questions.

At the end of the course students should be able to understand and explain - basic properties of natural substances (aminoacids, carbohydrates, lipids, nucleic acids, proteins, enzymes and coenzymes, their properties and structure, active site), thermodynamics, enzyme kinetics, inhibition, allostery, regulation of enzyme activity.

  • Qualitative and quantitative determination of carbohydrates. Qualitative and quantitative determination of aminoacids. Quantitative determination of proteins. Isolation, analysis and determination of nucleic acids. Separation methods. Determination of enzyme activity. pH effect on enzyme reaction. Enzyme kinetics. Inhibition of enzymes. At the end of the course students should be able to understand and explain - basic properties of natural substances(aminoacids, carbohydrates, lipids, nucleic acids, proteins, enzymes and coenzymes, their properties and structure, active site), thermodynamics, enzyme kinetics, inhibition, allostery, regulation of enzyme acitivity.

Course objectives
The objective of the laboratory course are studies of basic properties of compounds important for biochemistry, and methodological approaches in the study and analytical application of enzymes. Content of the first part includes qualitative and quantitative reactions of carbohydrates, lipids, amino acids, proteins and nucleic acids. The second part is oriented to enzyme kinetics and further studies on properties of free enzymes as well as enzymes which are immobilized in cells.

  • Qualitative reactions of carbohydrates. Carbohydrate thin-layer chromatography.
  • Isolation and identification of lipids from the egg yolk.
  • Separation of lipophilic pigments using adsorption chromatography.
  • Chemical reactions of amino acids and proteins. Paper chromatography of amino acids.
  • Isolation of nucleoprotein from spleen.
  • Gel permeation chromatography in separation of biomolecules.

At the end of the course students should be able to understand and explain - basic properties of natural substances(aminoacids, carbohydrates, lipids, nucleic acids, proteins, enzymes and coenzymes, their properties and structure, active site), thermodynamics, enzyme kinetics, inhibition, allostery, regulation of enzyme acitivity.
