- During practical courses, students will extract and quantify the pigments, glutathione and antioxidative enzymes from different plant material. The influence of stress conditions on analyzed compounds in plants will be demonstrated in easy experiments. Students will acquire basic methods of plant sample handling procedure and preparation. Last, but not least, students will learn advance operating of spectrophotometer and HPLC.
- Syllabus
- 1. Introduction. Safety in lab, requirement for successful termination.
- 2. Pigments extraction from plant sample. Different extraction reagents.
- 3. Thin-layer chromatography. Pigments absorbtion spectra.
- 4. Preparation of standards from commercial prepared standards. Pigments standards analysis on HPLC.
- 5. Evaluation of pigments chromatogrammes. Establishing of experiment Light conditions during the plant growth
- 6. Sample preparations. Freeze-drying. Grinding. Rules during sample preparation.
- 7. Study of the influence of low temperature on antioxidative enzyme activities. - I. catalase with oxygen electrode.
- 8. Study of the influence of low temperature on antioxidative enzyme activities. – II. ascorbate peroxidase by spectrophotometer. Determination of protein amount by Bradford method.
- 9. Sampling from the first experiment. Quantification of pigments spectrophotometrically.
- 10. Pigments extractions from the first experiment. HPLC analysis.
- 11. Extraction of glutathione from the first experiment and determination of its amount and redox state by HPLC.
- 12. Extraction of ascorbate and determination of its amount and redox state by HPLC.
- 13. Chromatogrammes analysis.
