Znalost molekulární struktury a funkce prokaryotického a eukaryotického genomu. Znalost základních mikrobiologických technik jako kultivace bakterií, stanovení titru bakteriální suspenze. Znalost výpočtů složení chemických roztoků a jejich příprava. Předpokládá se předchozí absolvování základních cvičení z anorganické a organické chemie a mikrobiologie. - Course Enrolment Limitations

- Course objectives
- At the end of the course students should be able to competently analyse the genetic material including chromosome DNA and solve experiments with plasmids originating from the bacteria.
- Learning outcomes
- Student will be able to:
isolate plasmid DNA of sufficient quality and quantity;
analyze isolated genetic material by basic methods of molecular biology such as restriction endonuclease cleavage and ELFO;
prepare sufficient amount of PCR amplicon of the gene of interest;
create the construct of the plasmid vector and the insert of the gene of interest and perform the transformation of competent cells;
use theoretical knowledge of other methods and principles in subsequent laboratory practice - Syllabus
- This course include the practical tasks which serve to acquire the basic methods in the molecular biology, primarily, isolation and manipulation with nucleic acids. Preparation, solutions useful for molecular methods: Isolation of plasmid DNA Concentration and purity of DNA Restriction splitting, ELFO Ultracentrifugation
