- Course objectives
- The practical course accompanies the lecture Mechanisms of cell death, significance, methods. Within the two six-hour-blocks students will have an opportunity to practically learn and perform several selected modern methods of apoptosis detection using flow cytometry and microscopy. Due o the complex innovation of this course in 2015, several new flow cytometry-based methods of the cell death detection have been introduced and optimized for the effective realization within the practical courses. Specifically, students will have an opportunity to learn multiparametric analysis of the activation of selected specific molecules (active caspase-3) and events (changes in plasma membrane summetry, phosphatidyl serine translocation) involved in cell death regulation. Moreover, these will be studied simultaneously in combination with analysis of other important cellular processes such as cell death progression (total DNA content, propidium iodide staining), DNA damage (gamma-H2AX phosphorylation) and cell viability (specific probes). Therefore, the students will benifit not only from learning to detect the „classical“ cell death parameters, but also to study them in context of other crucial processes affecting the final cell fate. The innovation of the practical course represents important improvement in complex study of the cell death and correct interpretation of the data obtained. Finally, this will enable students to get better and more relevant view of the complex cell death regulation. They will be provided with information how to evaluate the results obtained, interprete the data, and get practical recommendations about the application of individual methods, their advantages, disadvantages, and limits.
