- Course objectives
- The main objectives of the course is to become oriented in the enviroment of chemical laboratory and its equipment, to acquire simple laboratory procedures and techniques, to practice common separation procedures and become familiar with handling common laboratory machines.Students will learn to handle laboratory data into the lab reports, to solve the problems and derive conclusions.
- Learning outcomes
- Students will be able to understand basics of general, inorganic and organic chemistry to the extent necessary for the further study of biochemistry. • They will be able to apply general physicochemical laws to processes in living organisms. • They will be able to describe the chemical properties of the most important biogenic elements and essential compounds of importance in medicine, nutrition and toxicology. They will be able to describe and discuss the properties and reactions of organic and bioorganic compounds important for the understanding of biochemical processes.
- Syllabus
- Rules of study and directions for laboratory work General safety guidelines. Handling chemical substances – main regulations. Measuring of liquid volume. Advantages and disadvantages of plastic ware. Clearing of glass and plastics. Heating, cooling, freezing. Thermostats. Weighing, types of weights. Demonstration: Various types of weights, handling with them. Demonstration: Volumetric glassware Demonstration: Pressure cylinders and manipulation with them Measuring of small volumes of liquids, calibration of pipettors, expression of accuracy and precision. Demonstration: using of pipettors, principles of precise pipetting Gravimetric calibration of micropipettors. Verification of precision of repeated pipettting. Preparation of solutions with given concentration. Photometric verification of concentration Filtration, dialysis, ultrafiltration and microfiltration , centrifugation. Demonstration: Using of various centrifuges, replacement of rotors, calculations. Preparation of saline solution. Purification of Al2O3 for chromatography Dialysis of starch and podium chloride. Separation of precipitated proteins by centrifugation. Distillation, extraction. Extraction of drugs from urine. Demonstration : Extraction on solid phase. Preparation of redistilled water. Demonstration: Extraction on Soxhlet apparatus Demonstration: Using of vacuum evaporator Measuring and calculation pH of solutions of acids, bases and salts, pH meters a indicator strips. Preparation of volumetric solutions. Acid-base titrations, neutralization titration curves.. Determination of NaOH concentration using volumetric solution of oxalic acid. Determination of HCl concentration. Construction of titration curves of hydrochloric and acetic acid. Solution of buffers and their preparation. Preparation of hydrogenphosphate buffer of various pH values. Titration of a buffer. Determination of bufferin kapacity. Ionic reactions. Ions in dribling water. Solubility of compounds. Ion-selective electrodes. Detection and quantitative determination of nitrates, nitrites and amonium cations in water. Properties and reactions of phosphate ions. Properties and reactions of carbonate ions. Liquid chromatography – types of, basic methods, gas chromatography. Separation of potassium dichromate and blue-dextran by gel chromatography. Adsorption chromatography on the thin layer of silica gel. Demonstration: High pressure liquid chromatography. Demonstration: Deionization of water, preparation of ultra pure water. Spectrophotometry Spektrofotometric determination of salicylic acid Simple test-tube reaction of organic compounds. Determination of ethanol in blood theory, calculations. Reduction properties of monosaccharides and L-ascorbic acid Color reactions of amino acids and proteins. Properties of proteins, quantification of proteins. Determination of isoelectric point of cassein Solubility of proteins, protein precipitation.

- Teaching methods
- The course is realized in laboratories. Students are doing simple experiments according to the instructions in the textbook.On the base of results they prepare the lab-reports.
- Assessment methods
- Conditions for giving credit:Full attendance in all lessons (making up all missing and justified lessons), completion of all lab reports and handing them to the teacher till given date.Twelve test are writen during the semester. Student will recieve course unit credit, if he has at least 70% of points. The other students will write the credit test.This test has 30 questions and student must obtain at least 12 points. The test can be once time repeated.
