- Course objectives
- At the end of the course, students should be able to: - understand the problems of trace analysis - choose system of work in laboratory for trace analysis - outline parameters important for environment of clean laboratory - compare capacities for methods for preconcentration of trace elements and separation of matrix - appraise advantages of flow analysis - propose suitable analytical procedure and method for practical application
- Syllabus
- 1. Special problems in work with small sample quantity and with extremely low concentration. Microgravimetry, microtitrations.
- 2. Instrumental microanalysis, ultramicroanalysis.
- 3. Importance of trace analysis for practice. Laboratory for trace analysis, its equipment and rules for users.
- 4. Chemical vessels. Reagents, their purification and storing. Purity of chemicals and water.
- 5. The sample, its homogeneity and storing. Contamination and analyte loses in the chemical operations.
- 6. Decomposition of inorganic materials, pressure systems, bomb with teflon and quartz vessels.
- 7. Decomposition of biological materials, wet ashing, oxidising mineral acids at normal pressure,high pressure digestion, microwave oven, Fentons reagent. Photolytic decomposition.
- 8. High temperature dry ashing, muffle furnace, radio frequency plasma.
- 9. Separation of trace and matrix elements. Volatilization.
- 10. Selective dissolution, liquid-liquid extraction, sorption, ion exchange.
- 11. Flow injection analysis, zone dispersion, FIA systems. Preconcentration of elements on chelating sorbents. Flow injection extraction system, segmentor, phase separator.
- 12. Freezing and zone melting. Precipitation of trace elements, carrier precipitation and flotation.
- 13. Electrochemical deposition, electrodeposition on solid electrodes and mercury cathodes, flow and static arrangement, Coupling of deposition with AAS. Blank and its importance in trace analysis.
- 14. Local analysis of the surface, LA-ICP-OES.

- Teaching methods
- Education is performed as lectures with PowerPoint presentation. Understanding of solution of trace analysis problems, preconcentration methods, flow analysis, data evaluate and the use in practical analysis is emphasized.
- Assessment methods
- Final assessment (at the end of semester) is by oral examination. The exam consists in six questions, which require description and explanation of asked topic.
