- Course objectives
- At the end of this course, students should be able to:
describe the development of angiosperm reproductive organs;
understand the processes of pollen-pistil interactions;
explain the process of double fertilization in angiosperms and to interpret seed development. Regulation of above mentioned processes will be illustrated on selected plant models. - Syllabus
- Seed as a start of a new life cycle: angiosperm seed structure (testa, endosperm, aleuron layer, embryo: embryonal axis, cotyledones, hypocotyl), seed quiescence and dormancy, seed germination, seedling structure.
- Mitosis - nuclear division: Cell cycle, cytoskeleton formation during cell cycle, polarity, asymmetric divisions, squash methods, pretreatments for karyology, cell cycle synchronization.
- Morphology of the vegetative stem apical meristem (structure, stem cells, leaf primordia and leaf meristems, leaf differentiation); root apical meristem (quiescent centre, root cap, columela).
- Megagametogenesis, gynaeceum, pistil, angiosperm ovule structure, clearing methods of plant ovules, observation of cleared ovules by means of Nomarski DIC.
- Microsporogenesis and microgametogenesis, androeceum, stamen: anther and filamentum, structure of the anther wall, tetrades, types of pollen grains, differential staining of pollen grains, determination of pollen viability.
- Pollen grain germination in vitro, phase contrast microscopy.
- Pollen - pistil interaction: pollen tube growth, callose plugs, decolorised aniline blue staining, fluorescence microscopy in UV light.
- Cryostat sections in embryology - fixation, cryoprotection, cryo-sectioning (cryostat), enzyme histochemistry, histochemical reactions of hydrolytic enzymes.
- Histological techniques in plant embryology I. - fixation of isolated organs.
- Histological techniques in plant embryology II. Dehydration of samples, paraffin and resin embedding, microtome sectioning, adhesion of sections, de-waxing of sections.
- Histological techniques in plant embryology III. Successive double staining of sections, dehydration and Eukitt mounting.
- Documentation of prepared microscopic preparations - drawings, measurements, micrographs, automatic analysis of images.
