- Knowledge of Analytical Chemistry on a level of introductory course Knowledge of high school Physics. The course is given in Czech or English - as requested by students. The final oral examination will be in Czech or English.

- Course objectives
- Student will understand fundamentals of mass spektrometry including ionization methods, mass analyzers, ion detectors and selected applications, e.g. ionization mtechniques MALDI and ESI, modern mass snalyzers time-of-flight, ion traps and orbitrap, identification and quantification of proteins.
- Syllabus
- 1. A Brief Historical Perspective: Overview of Techniques and Technology. Basic Concepts of MS (resolution, sensitivity).
- 2. Ionization Techniques and Sample Introduction: Electron Impact (EI). Chemical Ionization (CI). Glow Discharge. Inductively Coupled Plasma (ICP). Fast Atom Bombardment (FAB). Secondary Ion Mass Spectrometry (SIMS). Thermospray (TSI). Electrospray (ESI). Laser Desorption (LD). Plasma Desorption (PD). Matrix-Assisted Laser Desorption/Ionization (MALDI). Coupling of Separation and MS (on-line, off-line, chips)
- 3. Mass Spectrometers: Ion Optics. Simulation of Ion Movement (Simion). Energy Analyzers. Magnetic Sector. Quadrupole Mass Analyzer. Ion Cyclotron Resonance Fourier Transform Mass Spectrometer (FT-ICR-MS). Ion Trap (IT). Linear Trap (LT). Orbitrap. Time-of-Flight Mass Spectrometer (TOFMS). Collision-Induced Dissociation (CID). Tandem Mass Spectrometry (MS/MS). Vacuum Principles & Techniques. Detectors & Detection Electronics.

- Teaching methods
- Lectures and discussion in the end of the semester. Visit of a mass spectrometry laboratory, demonstrations of MS hardware.
- Assessment methods
- Final oral examination (in Czech or English). The exam consists of three questions on the following topics: 1. ionization techniques, 2. mass analyzers, 3. applications. Fifty percent of correct answers is needed to pass.
