Course objectives
The aim of this practical laboratory course is to give the possibility to observe bacteria in their morphological diversity and to study the structures of bacterial cells. The course could give basic practical knowledge of methods of microscopic slides preparation, microscopy and image acquisition.
Syllabus
  • Morphology of bacterial cells and colonies, basic staining methods. Techniques of work with light microscope, care, removing of simple problems. Microphotography, digital photography, the use of video-cameras in image acquisition in microbiology, computer image analysis. Determination of the size of bacterial cells and their morphological structures(the use of ocular micrometer and software for image analysis). Gram staining. Acid-fast bacteria, Ziehl-Neelsen staining, mycobacteria. Giemsa staining. Endospores and sporulation of bacteria. Phase contrast and Nomarski diferential interference contrast. Capsules, sheaths and slimes of bacterial cells. Movement of bacterial cells. Technique of slide cultures. Actinomycetes and methods of their morphology observation. Measurment and comparing of proportions of selected groups of microorganisms. Staining of cell wall, nuclear material and reserved compounds in cells of bacteria and yeasts.
