- Course objectives
- Gene engineering, its history and perspective. In vitro mutagenesis as a method for changing genes, random mutagenesis, primer extension methods, cassette mutagenesis, PCR methods of site-directed mutagenesis. Protein engineering. Optimizing the expression of cloned genes, factor affecting the gene expression in a new host. Cloning in Gram-positive bacteria. Eukaryotic vectors, selectable marker for eukaryotic cells. Transfer of recombinant DNA into eukaryotic organisms. Cloning in yeast. Gene transfer to plants. Cloning in animal cells. Transferring genes into animal oocytes, eggs, embryos and specific animal tissues. Introduction of foreign genes into mice. Transgenic organisms. Gene therapy. Application of recombinant DNA to biotechnology. The impact of recombinant DNA technology.
