Course objectives
At the end of this course, students should be able to perform methods of DNA cloning, preparation of molecular probes, use polymerase chain reaction for modification od DNA ends and prepare DNA samples for sequencing.
Syllabus
  • 1. Isolation of Sau3AI and Sau96I restriction endonucleases.
  • 2. Cloning in pUC plasmid vectors (defosforylation of DNA ends by alcalic phosphatase, ligaion of vector and foreign DNA by T4 DNA ligase, transformation of competent cells, identification of recombinant clones on ampicillin plates, restriction analysis and construction of restriction maps of recombinant plasmids).
  • 3. DNA hybridization (non-radioactive labeling of DNA by digoxigenin, Southern bloting, hybridization with labeled probe, immunologic detection).
  • 4. Modification od DNA ends by PCR, cloning of PCR products.
  • 5. DNA sequencing.

Teaching methods
The course is taught in one week block in research molecular biology laboratories of the department. Students perform practical experiments according to the instructions of teacher and according to printed manuals.
Assessment methods
The course is completed after work out an elaborate and pass a writen test.
