- The course is offered to students of any study field.
- Course objectives
- Modification of biomolecules based on conjugation reactions, introduction of labels and immobilization on surface of polymers and sensing surfaces is used in many current research procedures, bioanalytical assays and therapeutic methods. Initially, the suitable surface target groups of proteins, nucleic acids and saccharides will be mentioned. Next, suitable reagents for conjugation and labelling (enzymes, fluorophores, biotinylation, liposomes, nanoparticles). Methods for activation of surfaces for binding of biomolecules. Characterisation of bioconjugates. Typical applications (hapten-carrier conjugates, labelled antibodies, affinity chromatography, immobilised enzymes, protein and DNA biochips.
- Syllabus
- 1. Modification of proteins and peptides, side chains of amino acids, prosthetic groups.
- 2. Modification of nucletides, DNA, RNA. Modification of (poly)saccharides, glycoconjugates.
- 3. Insertion of additional reactive groups into biomolecules. Basic chemical reactions for conjugation.
- 4. Bioconjugations, homo/heterobifunctional reagents. Cleavable links.
- 5. Fluorescent labels, fluoresceins, rhodamins, coumarins, FRET.
- 6. Biotinylation, chelates, oligohistidine tags, boronate complexes. Radiolabels, nanoparticles, liposomes.
