Course Enrolment Limitations
The course is offered to students of any study field.
Course objectives
Students will gain theoretical knewledge about modification of biomolecules based on conjugation reactions, introduction of labels and immobilization on surface of polymers and sensing surfaces. They will obtain overview of reagents for conjugation and labelling (enzymes, fluorophores, biotinylation, liposomes, nanoparticles).
Learning outcomes
Students will learn methods for conjugation of biomolecules with labels as enzymes, fluorophores and nanoparticles. They will manage principles of immobilization techniques for preparation of affinity supports and surfaces for purification and bioanalytical applications.
Syllabus
  • 1. Modification of proteins and peptides, side chains of amino acids, prosthetic groups.
  • 2. Modification of nucleotides, DNA, RNA. Modification of (poly)saccharides, glycoconjugates.
  • 3. Insertion of additional reactive groups into biomolecules. Basic chemical reactions for conjugation.
  • 4. Bioconjugations, homo/heterobifunctional reagents. Cleavable links.
  • 5. Fluorescent labels, fluoresceins, rhodamins, coumarins, FRET.
  • 6. Biotinylation, chelates, oligohistidine tags, boronate complexes. Radiolabels, nanoparticles, liposomes.
  • 7. Enzyme labels.
  • 8. Immobilization of biomolecules for affinity chromatography.
  • 9. Activation of inert sensing surfaces - gold, platinum, graphite.
  • 10. Characterization of bioconjugates.
  • 11. Hapten-carrier conjugations.
  • 12. Immunosensors, sensing microarrays and DNA biochips.
