Course objectives
The course is follow-up of the course in Bi6120 Plant explants, in which students are familiar with various methods of in vitro cultivation. Expands knowledge towards applications of biotechnology for obtaining natural products available for pharmaceutical use ("Pharming"), food and cosmetic purposes, the preparation of edible vaccines, remediation. Part of the course will focus on GMOs, higher plants with resistance to pests, diseases or adverse external influences. From a methodological point of view, mainly an suspension and submerged culture, elicitation, bioconversion, encapsulation and cryopreservation.
Learning outcomes
At the end of this course, students should be able:
- work in sterile conditions used in in vitro plants cultivations;
- prepare media for in vitro plant cultivations;
- use suitable method of in vitro plant cultivation in plant biotechnology (e.g.: callus, suspension cultures);
- suggest and perform selection of transgene plants;
- describe and use some basic molecular techniques (DNA isolation, electrophoresis, PCR);
- describe methods used in preparation of trasgene plants;
- describe and use methods of plant cultivation in bioreactors;

  • 1. Preparation of cultivation media;
  • 2. Work under sterile conditions, sterile seeding on selective medium;
  • 3. Subcultivation of in vitro plants;
  • 4. Transfer of plants in RITA bioreactors;
  • 5. Callus cultures;
  • 6. Initiation and maintenance of suspension cultures;
  • 7. Transformation by Agrobacterium rhizogenes;
  • 8. Organ cultures;
  • 9. Initiation and maintenance of green algea in bioreactors;
  • 10. Plant DNA extraction;
  • 11. SSRs method for testing of somaclonal variability and different plant varieties.
