- Course objectives
- The aim of the course is to acquire basic bioengineering knowledge enabling to understand and control the processes of biochemical engineering for the needs of biotechnological research and development and for application in industrial practice.
- Learning outcomes
- Student will be able to: - be knowledgeable about the main findings in biochemical engineering under laboratory and industrial scale; - understand basic processes and equipment in biotechnology using microbial and other cell cultures and enzymes; - control the basics of bioprocess kinetics to describe and model its course in bioreactors.
- Syllabus
- Bioengineering - biochemical engineering in controlling biotechnological processes. Biocatalysts in biotechnology, from enzymes to microbial, plant and animal cells, relationships between enzyme and cell kinetics. • Bioreactors as universal cultivation and production devices. Main features and equipment of bioreactors. • Scale up. Development of biotechnology process from laboratory to industrial scale. • Specificity of bioreactors and culture devices for plant and animal cultures. • Processes in bioreactors. Stirring and aeration, determination of optimal parameters in terms of economics and bio-process biochemistry. • Critical aeration parameters in terms of substrate oxidation and overall bioprocess kinetics. • Sterilization of materials, nutrient media and air. • Biomass production. Batch and fed-batch cultures, bioprocess constants. • Kinetic models of biomass growth, substrate consumption and product formation. Cell death and autolysis. Application of a suitable model. • Continuous cultivation. Steady state, determination of the kinetic parameters under specific conditions of continuous process. • Use of the batch process conditions to determine the optimal parameters of continuous culture. • Bioreactors with immobilized biocatalysts. Application of enzymes and cells, kinetics of product formation. • Bioprocess monitoring, separation of biomass and products.
