- Course objectives
- Passing through this course, students are able to:
apply basic methods and techniques of plant cell, tissue and organ culture in vitro;
propose in vitro methods suitable for plant multiplication, breeding and production;
estimate efficiency of the selected method for specific plant material on the base of visible symptoms. - Learning outcomes
- At the end of this course, students should be able to:
- work in sterile conditions of in vitro cultures;
- prepare culture media and sterilize them;
- create and maintain different in vitro cultures;
- transfer plants from in vitro to glasshouse. - Syllabus
- 1. Conditions of in vitro Culture, Laboratory Design.
- 2. Media Preparation and Sterilization.
- 3. In vitro Vegetative Propagation: Micropropagation by Proliferation of Axillary Buds, Adventitious Shoot Proliferation; Initiation of Primary Culture, Subculture. Transfer to ex vitro conditions.
- 4. Callus Culture: Callus Initiation and Maintenance, Subculture, Organogenesis from Callus.
- 5. Meristem Culture: Apical and Axillary Meristem Isolation and Culture.
- 6. Embryo Culture: Isolation and Culture of Young and Mature Zygotic Embryos. 7. Somatic embryogenesis: induction of somatic embryos from different materials 8. Ploidy reduction: androgenesis: initiation of isolated anthers culture
- 9. Cryopreservation: Alginate Encapsulation/Dehydration of isolated axillary potato meristems.
- 10. Fungi Culture in vitro: Suspension Culture of Wood-Destroying Fungi.
