- Course objectives
- The goal of practical exercises in general microbiology is to teach students basic methods of work in microbiological laboratory (methods of sterile work, isolation, seeding and cultivation of microorganisms, preparation of microscopic preparations and work with microscope, determination of number of bacterial cells in different types of samples, yeasts and microscopic fungi).
- Learning outcomes
- Learning outcomes of the course unit will be practical knowledge and ability of students to use microbiological methods in the practice.
- Syllabus
- Introduction, organization of exercises, safety of work in microbiological laboratory. Preparation of nutrient media and their sterilization. Isolation, seeding and cultivation of microorganisms. Microscopic observation of bacterial cells (native and fixed slides, Gram staining, work with microscope and image analysis). Determination of the total number of viable of bacterial cells by a plate method. Bacteria in water: basic microbiological analysis of the water. Determination of concentration of antibiotics by microbiological methods. Determination of bacterial sensitivity to antibiotics. Basic identification of bacteria, biochemical and physiological tests. Yeast, native slide, vital test, slide culture, cell size measurement. Detection and isolation of some soil microorganisms (cellulolytic activity, Azotobacter, Clostridium), microscopic fiber fungi. Assessment of the presence of microorganisms in the soil. Negative staining. Staining of Azotobacter sp. Observation of bacterial spore. Microscopic fibrous fungi: Credit final test.

- Teaching methods
- At the beginning of each practical class, the students are acquainted with the content of the class in the form of explanation and demonstrations, then perform the assigned practical task. They draw up a protocol from each class.
