- Course objectives
- Classical organic analysis = characteristic physical properties of organic compounds, elemental analysis of organic compounds, analytical identification and determination of important functional groups of organic compounds, mainly by colour reactions in solution. Interpretation of spectra of simple organic compounds obtained by instrumental methods: UV-VIS, IR, NMR, MS.
- Syllabus
- Characteristics of analysis of organic compounds. The methodology of the analysis, development and trends. Determination of physical constants: melting and boiling point, refractivity, optical activity, partition coefficients of extraction, molecular weight, spectral constants. Elemental analysis. Sample decomposition, detection and determination of C, H, O, N, Cl, Br, I, S, P, F. Automatic methods of elemental analysis. Functional groups identification: classes of dissolution, classification reactions for carbohydrates, alcohols, phenols, aldehydes, ketons, carboxylic acids, acid anhydrides, esters, amides, imides, ethers, epoxides, peroxides, amines, nitriles, isonitriles, nitro-, nitroso-, azo-, azoxy-, hydrazo- and diazo- compounds, thiols, sulphonic and sulphinic acids, halogenic compounds. Determination of organic compounds based on their functional groups reactions. Determination of active hydrogen, determination of hydroxy-, carbonyl-, carboxyl-, amino-, nitro-, nitroso-, thio- a halogeno-compounds. Liquid-liquid extraction, thin layer chromatography (TLC). Instrumental methods of organic analysis - principles (gas chromatography (GC), liquid chromatography (LC), spectroscopy, optical rotation dispersion (ORD), nuclear magnetic resonance (NMR) and mass spectrometry (MS)). Spectra interpretation in organic analysis UV-VIS spectroscopy: sigma and pi orbitals, chromophores, allowed and disallowed electron transitions, Woodward-Fieser rules, examples. Infra red (IR) spectroscopy: models of fundamental and normal vibration, procedure of IR spectra interpretation with tables of characteristic vibrations. Nuclear magnetic resonance (NMR): chemical shift, first order spectra and their interpretation, spin interaction constant, decoupling, examples. Mass spectrometry (MS): specifics of the spectra, determination of summary formula from the natural nuclides distribution, fragments analysis of organic molecules, ionradical, molecular ion, empirical rules, carbcation stability, nitrogen rules, examples.
