Basic knowledge from molecular biology of the Prokaryotes. Basic techniques of microbiology. - Course Enrolment Limitations
- The course is only offered to the students of the study fields the course is directly associated with.
The capacity limit for the course is 12 student(s).
Current registration and enrolment status: enrolled: 0/12, only registered: 0/12 - fields of study / plans the course is directly associated with
- Molecular Biology and Genetics (programme PřF, M-BI)
- Molecular Biology and Genetics (programme PřF, N-BI)
- Course objectives
- Practical course focused on molecular methods for analysis of procaryotic genome and basic methods of bacterial genetics. At the end of this course, students should be able to design and perfotm basic laboratory experiments focused on genetic studies of Escherichia coli and selected species of Gramm-positive cocci.
- Syllabus
- 1. Pulsed-field gel electrophoresis of bacterial genomic DNA (preparation of samples, digestion with restriction endonuclease SmaI, PFGE).
- 2. Isolation of spontaneous mutants of Staphylococcus aureus resistant to antibiotics on gradient agar plates.
- 3. Fluctuation test and estimation of mutation rate.
- 4. Conjugation of Escherichia coli.
- 5. Isolation of auxotroph mutants of Staphylococcus aureus. Replica plating.
- 6. Typing techniques used for epidemiological, and phylogenetical purposes, AP-PCR, RFLP.
