Prerequisites
The course is destined for students interested in microbiology and biotechnology. Enrolling in the course requires as a prerequisite the examination in the course Molecular Biotechnology (Bi7430)

Course objectives
At the end of this course, the students will be able to understand of the principes of methods and processes used at the isolation of biotechnological products. Students will demonstrate knowledge of the use of methods for the solving of practical probleme.
Syllabus
  • Stechiometry of microbial growth. Material balances.
  • Fluids properties. Fluid transfer in piping.
  • Bioreactors support systems and processes. Systems with immobilized cells and enzymes.
  • Influence of solvents on enzyme activities.
  • Methods of product isolation. Mechanical, chemical and enzymatic cell disruption. Filtration. Centrifugation.
  • Precipitation. Salting out. Denaturation and renaturation of proteins.
  • Liquid-liquid extraction. Aqueous two-phase systems. Types of extractors and their design.
  • . Membrane separation processes. Large scale chromatographic purification of proteins.
  • Drying. Storage and stability of proteins. Validation of production quality.

Teaching methods
The teaching methods make use of presentations on stencils. For the explanation of problems are used practical examples and calculations.
Assessment methods
The course is closed with an oral exam. The student answers 3 questions in the exam. The overall result of the exam is evaluated by the examiner.
