- Course objectives
- An advanced lecture. Enzymes preparation and storage. Enzyme nomenclature. Enzyme structure and kinetics. Enzyme analysis. Use of enzymes in biotechnology. Bioanalytical applications of enzymes. Technics of enzyme immobilization. Enzyme biosensors and enzyme reactors. Enzymes in immunochemical techniques.
- Syllabus
- 1. Intial information on enzymes. Basic terms. Enzyme activity. 2. Methods for measuring enzyme activity - optical and electrochemcial, suitable substrates. Examples of measurement for most important enzymes. 3. Isolation of enzymes, purification procedures, chromatography. Commercial sources of enzymes. Enzyme structure characterisation. 4. Mechanisms of enzyme catalysis. Basic principles, typical examples. 5. Kinetics of enzyme reaction with substrate, parameters vmax (vlim) and Km including methods for determination. Software for enzyme kinetics, examples of use. 6. Reactions with multiple substrates, classification, different mechanisms. Inhibitors, types, inhibition constants, kinetic studies. 7. Effect of external factors on activity (pH, temperature, ionic strength, viscosity). Cooperative effects in enzyme action. 8. Bioanalytical application of enzymes. Enzyme assays in clinical analysis. 9. Enzyme-based biosensors, measuring systems, examples of applications. 10. Immobilization of enzymes, enzyme reactors. Entrapment inside polymers, covalent attachment on beads and sensing surfaces. 11. Enzymes in immunochemical techniques, ELISA. Enzyme labels and methods for preparation of enzyme conjugates. 12. Industrial application of enzymes. Information sources on enzymes on internet.
