- Course objectives
- At the end of the course students should be able to understand principles of work with bacterial and yeasts cells and bacteriophages and to understand chosen tasks, to solve various problems they can face in bacterial physiology; to use set of chosen techniques used in physiological laboratories; to work with growth characteristics of bacterial cells and phages, to understand influence of temperature and UV on lysogenic cells and phages; to prepare crude enzymatic preparation and to test its enzymatic activity and test physiological properties of industrial strains of yeasts. Students will learn to evaluate experimental data acquired, to interpret these results and to deduce conclusions from them. In case of unexpected results they will try to identify possible mistakes/problems. Students will produce a written report from acquired results as preparation for publication in scientific journals.
- Syllabus
- Determination of bacterial proteins and dry-weight. Bacterial growth curve in static culture (using plate counts). One-step growth curve of polyvalent and virulent staphylopage. Release of bacteriophage from lysogenic culture by UV induction. Heat inactivation of a bacteriophage. Determination of dehydrogenase activity. Preparation of crude enzymatic product(HEP) with phosphatase activity. Determination of phosphatase activity of native cells and HEP. Influence of heavy metals on bacterial growth (Bioscreen)and protein production. Flocculation of yeasts.

- Schemata pro jednotlivé úlohy poskytne vyučující. Laboratory protocols are provided by teacher.

- Teaching methods
- Topic of each practical is at first explained and demonstrated to students. Then students are solving the practical task by themselves. The outcome of each practical is written report.
