C8980 Protein preparation
Basic knowledge of biochemistry and/or molecular biology is a prerequisite for registration for the course. Students must pass this course (C8980 Expression and purification of proteins) before registration for course C8980c.

Course objectives
After completion of the course, the student will posess basic practical knowledge of how to express and purify recombinant proteins, will have experience with various expression systems and will be able to use different purification methods. During the course the student will be acquainted with the various methods of detection and is able to mark proteins for NMR analysis or crystallography.
Syllabus
  • 1. Strategy of cloning into expression plasmids. 2. Expression and purification of the human scFv produced in cytoplasm or periplasmic space. 3. Expression and purification of the plant protein (A. pET32 – thioredoxin - chaperon/B. pRSET – His.tag/ C. pET161DEST – Gate way technology – Lumio.tag/D. pET28 – without any tag.)
