Course objectives
The aim of the practical course is to familiarize the students with basic methods of current mycorrhizal research.
Syllabus
  • Mycorrhizal colonization of plant roots: review of commonly used methods, assessment of root colonization (chemical and microscopic methods), observation of typical mycorrhizal structures.

  • Isolation of spores of VAM fungi: isolation techniques, stain by Melzer's reagent, sample preparation, spore description and fungal species determination.

  • Mycorrhizal extraradical mycelium: extraction techniques (membrane filtration and inserted membrane technique), vital staining techniques, assessment of extraradical hyphal length (grid-line intersect method and image analysis).

  • Long-term cultivation experiment: the aim of the experiment vary with every course run.
